Job Summary
Reporting to the Principal Investigator, the Research Associate is responsible for the receipt, handling and analysis of samples submitted for diagnostic genetic testing, involving the isolation of DNA, amplification of appropriate regions of the genome and subsequent sequence analysis of the amplified DNA region to evaluate the presence of genetic variants. The incumbent is expected to use personal judgement in the development of priorities for test performance and must be familiar with, and follow closely, laboratory protocols developed with reference to the laboratory’s accreditation with the Institute for Quality Management in Healthcare (IQMH). The incumbent will produce diagnostic test reports that will be reviewed and signed by the Principal Investigator. This position requires the incumbent to work with human blood samples and may be require work with hazardous materials. The Research Associate will also perform other support duties including ordering supplies, and the cleaning and maintenance of basic laboratory equipment.
Job Description
KEY RESPONSIBILITIES:
- Follow detailed protocols for the receipt, extraction and testing of DNA samples submitted from referring hemophilia and genetics clinics Canada-wide.
- Prepare reagents for the extraction of DNA from submitted human blood samples and conduct experiments to confirm the integrity and amount of DNA obtained from the extraction process.
- Conduct studies to amplify regions of the human genome on extracted DNA samples. The regions of the genome to be amplified will be chosen based on the clinical history and requests submitted by the referring clinic.
- Perform experiments to ensure that amplification of the chosen DNA regions has progressed as expected. Send amplified DNA regions to the regional DNA sequencing facility for DNA analysis.
- Receive electronic records of the DNA sequences obtained by the regional laboratory and conduct computer-based analysis of the sequence to evaluate the presence of variant sequences that might result in a disease state.
- Obtain independent validation of the DNA sequence analysis through data sharing with a laboratory colleague.
- Compose a written DNA variant report that is submitted to the Principal Investigator for review and signature, before being transmitted to the referring clinician.
- Conduct regular reviews to ensure all laboratory equipment is functioning as expected. Clean and maintain lab equipment and perform preventive maintenance procedures. Correct operational problems where possible.
- Maintain detailed records (written and computer-based) of all samples received, the procedures undertaken for every sample, and the results of the DNA analysis.
- Submission of genetic test results to the Canadian Bleeding Disorder Registry (CBDR) where appropriate.
- Contribute to the laboratory’s performance to maintain its formal IQMH accreditation status. Follow the laboratory’s standard operating procedures and other detailed protocols as described in detail in the Quality Management manual of the laboratory. Follow the strict confidentiality and privacy regulations described in the laboratory’s IQMH protocols.
- Provide regular updates at team meetings.
- Encourage safety in lab and perform basic safety procedures, including proper hazardous waste and sharps disposal. Liaise with Physical Plant Services (PPS) and Environmental Health
& Safety (EH&S) when required.
- Perform administrative procedures including word processing, database maintenance, scheduling, and inventory/ordering of lab supplies.
- Undertake additional duties as required in support of the lab.
REQUIRED QUALIFICATIONS:
- Post-secondary degree in health related field, combined with several years (5+ minimum) relevant experience with molecular biology/molecular genetic techniques.
- Basic molecular biology skills, including handling of human blood samples, extraction of DNA, quantification of DNA, design of PCR primers, PCR amplification of DNA, analysis of amplified DNA, use of DNA variant software.
- Technical proficiency in the use of equipment such as PCR machines, incubators, and biosafety cabinets, and in use of precise instruments.
- Consideration may be given to an equivalent combination of education and experience.
SPECIAL SKILLS:
- Appropriate and safe handling of potentially biohazardous human samples.
- Attention to detail and ability to perform careful measurements and interpret basic experimental results.
- Organizational/analytical skills and ability to coordinate workflow. Judgement will need to be used to prioritize sample handling and test performance to ensure timely and correct test results are obtained.
- Care in handling dangerous materials, operating equipment, and preparing solutions or samples. Foresight in predicting problems and preventing them from occurring where possible.
- Computer skills to aid in database maintenance, DNA sequence analysis, and report generation.
- Strong interpersonal and communication skills to interact with a diverse audience, including various healthcare providers, when referring clients, through telephone and electronic means.
- Ability to work independently and collaboratively as a member of the laboratory.
- Ability to take initiative and to assess and establish priorities while working in an environment with multiple interruptions and strict deadlines.
- Strong team player who works well under pressure, with the ability to adapt to change and demonstrate flexibility and initiative. This includes the ability to respond to tight deadlines and conflicting priorities.
- Ability to learn quickly and adapt protocols as needs change.
- Flexibility in working hours to accommodate laboratory objectives.
DECISION MAKING:
- Decide on optimal experimental protocols and equipment to use in running an experiment and decide on steps to take when assigned a new project.
- Determine how to deal with breakdowns and errors. Decide if a problem can be resolved personally or if supervisor or equipment manufacturers should be involved.
- Determine best way to correct minor methodological deviations.
- Determine if additional priority input is required from the Principal Investigator.
- Direct inquiries to the proper person or supply the answer if appropriate.
- Prioritize time and determine most efficient use of personal resources within set of priorities outlined by supervisor.
- Coordinate test studies with members of the laboratory.
- Develop new DNA testing procedures based on the published literature and interactions with collaborating labs.
- Determine appropriate way to dispose of hazardous wastes within safety guidelines and react in an emergency to minimize damage.
